Materials and Methods

Serum specimen exhibition

The current case-control investigation was carried out from 2009 to 2010. An aggregate of 100 human serum specimens was amassed from various labs in Tehran, Iran. Antigen preparation

Tachyzoites of T. gondii were amassed from the peritoneal hole of mice that were infiltrated three days prematurely. Tachyzoites were rinsed with PBS three times, sonicated, and centrifuged at 12,000 g for 60 mins, and the supernatant was accumulated as soluble Ag. The polypeptide content was estimated by the technique of Bradford. The 96-well microtiter plates were covered with 5 µg/ml of the diluted polypeptide in a carbonate buffer (pH 9.6). Covered plates were placed at 4.C overnight, then rinsed and reserved at -20.C until usage.

Avidity Elisa

Microtiter dishes formerly covered with Toxoplasma Ag were rinsed three times with PBS plus 0.05%. Serum specimens were watered down 1/200 and put in (100 µl/well) on two rows of a dish (row A and row B), after gestation for forty-five min at 37.C; row B was rinsed three times with PBST, and row A was rinsed three times with the modified PBST buffer comprising 6 M urea and a fourth time with PBST.

After gestation and rinsing, the chromogenic substrate, o-phenylenediamine, was added. The Abs was read by an  ELISA reader at 492 nm. The avidity index was computed as the outcome of the Abs of wells washed with PBS-urea, split up by the Abs of wells washed with PBST, and multiplied by 100, based on the formula; AI=Abs(U+)/Abs(U-)×100.

Results

Among eighteen patients with acute toxoplasmosis, sixteen had an AI of fifty percent and quieter, and two showed a greater AI than fifty percent. The mean AI for these sixteen  sera was 44.39%. All of the sera were favorable for Toxoplasma-specific IgG and IgM Ab.
