In melanocytes and melanophores, there is an organelle called the melanosome that contains melanin. Specialized cells called melanophores, found in fish and amphibians, are loaded with hundreds of melanin-containing pigment granules known as melanosomes. These cells have only one purpose, which is pigment aggregation in the cell's centre or dispersion throughout the cytoplasm. The animal may affect colour changes necessary for camouflage and social interactions thanks to this alternate mode of pigment transfer.

What is Melanosome?

Melanophores move their pigment in response to external signals, which in frogs and fish are hormonal stimuli and neurotransmitters, respectively. In both situations, melanosome dispersion is brought on by increased intracellular cAMP levels, whereas aggregation is brought on by decreased cAMP. Uncertainty surrounds the regulatory processes that take place after these second messengers.

However, unlike melanophores, the pigment in mammalian melanocytes is delivered to the cell periphery for eventual exocytosis to neighbouring epithelial cells. Mammalian melanocytes also create melanosomes. Melanins are produced and stored in the tissue-specific "lysosome-related" organelles known as melanosomes found in pigment cells.

Formation

Melanosomes are produced through unique pathways from early endosomal intermediates, as is starting to become clear through studies of the trafficking and fate of their constituent parts. These investigations provide unanticipated insights into the production of multivesicular bodies and how compartmentalization controls protein refolding.

Why is this test recommended?

Melanins are intricate pigments that give mammals' skin, hair, and eyes colour and provide photoprotection against ionising radiation. Additionally, they play a role in the optic nervous system's development and control of retinal activity. Typically, copolymers of red and yellow phaeomelanin and black and brown eumelanin make up melanins. Through four physically distinct stages6, melanosomes mature within the melanocyte or the growing retinal pigment epithelial cell. Despite the absence of colour, the first two stages are distinguished by intraluminal proteinaceous fibrils, which start to form in stage I and are fully formed by stage II.

Melanin synthesis starts when the fibrous striations in ellipsoidal stage II melanosomes are fully developed. Melanins build up on the fibrils, causing them to thicken and turn black as they mature to stage III and stage IV, where all internal structure is hidden. Stage IV melanosomes are kept in the pigment cells of the eye, where they absorb stray light and possibly capture free radicals.

Stage IV melanosomes are transported from the cell centre to actin-rich dendritic terminals in epidermal melanocytes along microtubules.
