Abstract

Cancer treatment has changed dramatically as a result of the introduction of autologous chimeric antigen receptor T (CAR-T) cell treatments. The distribution of CAR-T cell treatment does, however, encounter difficulties, such as high costs, protracted production times, and manufacturing problems.

It has been attempted to overcome this by creating allogeneic CAR-T cells utilising donor-derived conventional CD4+ or CD8+ T cells (Tconvs), however this has proven difficult due to severe graft-versus-host disease (GvHD) and host immunological rejection. It has been demonstrated that CD3+CD4CD8 double-negative T cells (DNTs), a rare subset of mature T cells, can perform all of the necessary functions for an off-the-shelf cellular therapy, including scaling, cryopreservability, donor-independent anticancer function, resistance to rejection, and the absence of any observed off-tumor toxicity, such as GvHD.
