What does an amyloid protein test entail?

An acute-phase protein called serum amyloid. It is involved in the aetiology of atherosclerosis. The study's objective was to determine the level of SAA in RA patients' serum in relation to other inflammatory parameters and indicators of extra-articular involvement.

Amyloid Protein Identification

Ordering Guidance

  • Only patients with a primary diagnosis that has been established should have this test ordered. Order PATHC / Pathology Consultation or consult the Pathology Consultation Ordering Algorithm if a patient lacks a primary diagnosis

Important Information

1. An initial pathology report and medical history are needed.

2. It's also advised to send a quick letter of explanation or consultation.

Sample Required

  • Kit for pathology packaging
  • Formalin-fixed or B5-fixed, paraffin-embedded tissue block: Specimen Type

Collection Instructions: Don't send tissue slides that have been repaired. Only blocks of tissue with paraffin embedded can be tested.

Testing Method

Prior to mass spectrometry testing in every case with sufficient tissue, a preliminary Congo red stain is carried out to confirm positivity.

  • SS2PC / Special Stain, Group II, Other may occasionally be used to perform a different first Congo red stain at the pathologist's discretion.
  • This test will not be run and only the SS2PC will be charged if the stain is found to be amyloid-negative.
  • This test will be carried out and the SS2PC billing charge will be reimbursed if the amyloid stain is positive.

Normally, a pathology consultation is not necessary. Upon client consent, a PATHC / Pathology Consultation may be added if necessary in the event that the results of the amyloid subtyping do not match the clinical findings.

Clinical Details

The extracellular tissue deposition of misfolded proteins that causes end organ damage unites the hereditary and acquired disorders known as amyloidosis. The disease amyloidosis can be either systemic or localised. Despite the fact that many cases of amyloidosis are genetic, the majority are acquired due to underlying monoclonal B-cell/plasma cell cancer, as a result of age, or as a result of long-term chronic inflammation. Thus, particular amyloid proteins are linked to particular amyloid-related illnesses. These include serum amyloid A (SAA amyloid), transthyretin (ATTR amyloid), kappa or lambda immunoglobulin light chains (AL amyloid), and other unusual variants. Accurate identification of the proteins that make up the amyloid deposits is crucial since the therapy of patients with amyloidosis varies greatly depending on the many amyloid subtypes.
