Thrombotic thrombocytopenic purpura (TTP) is distinguished from other thrombotic microangiopathies by the ADAMTS13 test (TMAs). The ADAMTS13 deficiency pretest likelihood is determined by the PLASMIC score.

Either enzyme-linked immunosorbent assay, immunoblotting, or fluorescence resonance energy transfer (FRET) can be used to measure ADAMTS13 activity.

Describe ADAMTS13:

If the ADAMTS13 gene's activity is compromised due to a hereditary mutation or the emergence of an autoantibody, the buildup of ultra-large VWF multimers may result in microvascular thrombosis from platelet aggregation and trigger the syndrome of thrombotic thrombocytopenic purpura (TTP).

ADAMTS13 deficiency: What is it?

In humans, thrombotic thrombocytopenic purpura (TTP), a disorder marked by thrombocytopenia and hemolytic anaemia with microvascular platelet thrombi, is brought on by genetic or acquired

ADAMTS13 deficiency.

Results Interpretation:

  • Thrombotic thrombocytopenic purpura may be acquired (Upshaw-Schulman Syndrome) or inherited (ADAMTS13 levels less than 10%). (TTP).
  • A minor to moderate deficit of ADAMTS13 activity may be caused by several medical disorders. Recent plasma exchange therapy may increase the ADAMTS13 activity that has been noticed.

What Is ELISA?

Enzyme-linked immunosorbent assay, often known as EIA: Enzyme Immunoassay, is the fundamental assay technique used to identify and quantify antibodies, hormones, peptides, and proteins in the blood.

In the case of some infectious disorders, it is helpful to look at the body's antibody levels.

Since it separates non-specific and specific interactions that happen through serial binding to solid surfaces—typically a polystyrene multiwell plate—ELISA is a unique analysis when compared to other antibody assays.

ELISA test types:

According to the various strategies employed for binding between antigens and antibodies, ELISA tests can be divided into three categories, namely:

  • The antigen is coated to the microtiter well in an indirect ELISA.
  • Sandwich ELISA
  • Competitive ELISA: The antigen-antibody mixture is added to a Microtiter well that has been coated with an antigen.

ELISA Technique:

  • One of the simplest blood tests available is the ELISA.
  • It is quick and urgent and needs a patient's blood sample.
  • This is a description of the ELISA process in its entirety.
  • An antibody is bonded to a solid polystyrene plate that attracts or has an affinity for hormones, germs, and other antibodies.
  • This antigen-antibody mixture is added to an antigen-coated microtiter, which is then washed to eliminate any free antibodies.
  • It then contains a second, primary antibody-specific antibody that is typically enzyme-conjugated.
  • By washing the plate, free enzyme-linked secondary antibodies are eliminated.
