What is NOTCH3 Mutation Test?

Next-generation sequencing is used to detect single nucleotide and copy number variants in the NOTCH3 gene that are linked to cerebral autosomal dominant arteriopathy with subcortical infarcts and leukoencephalopathy (CADASIL) and other NOTCH3-related disorders.

The NOTCH3 gene codes for a protein with one end (the intracellular end) that remains inside the cell, a middle (transmembrane) section that spans the cell membrane, and an extracellular end that projects from the cell's outer surface. The NOTCH3 protein is known as a receptor protein because certain other proteins, known as ligands, attach (bind) to the extracellular end of NOTCH3. This binding results in the detachment of the NOTCH3 protein's intracellular end, known as the NOTCH3 intracellular domain, or NICD. The NICD enters the nucleus of the cell and regulates the activity (transcription) of other genes.
