Scared of Your Breast Lump? Diagnose Malignancy by Latest Technology RT-PCR

Breast Lumps are swelling or formation of solid muscular tissue in small region of breast may be below the breast or at the side of underarms, causing pain and discomfort with increasing in size day by day.

Breast Lumps are swelling or formation of solid muscular tissue in small region of breast may be below the breast or at the side of underarms, causing pain and discomfort with increasing in size day by day. These breast lumps can be benign or malignant and only found in women. According to the most recent reports about 30% of the women are diagnosed with breast cancer every year. This breast cancer or breast lumps can be the result of mutation of specific types of genes who promote growth and cell proliferation. The details behind this are discussed below with best diagnostic methods mentioned. Have a thorough read.

PIK3CA: The possible reason behind it

The official full form of PIK3CA is Phosphatidylinositol-4.5-bisphosphate-3-kinase Catalytic subunit Alpha (human). This PIK3CA is an oncogenic, protein coding 34 kb gene, which is also well known to us as PI3K-alpha, p110-alpha. This gene is comprised of catalytic subunits and has been reported to render oncogenic effects for colorectal cancers.

How does PIK3CA works in normal human body?

PIK3CA is the gene which encodes a subunit called p110alpha of protein phosphatidylinositol 3-kinase. This helps in cellular primary growth by triggering cell division, differentiation among them, and further proliferation.

Mutation of PIK3CA and its resultant defective functioning in human body

PIK3CA gene sometimes undergoes genetic changes, i.e., mutation at different levels, which causes intracellular trafficking that leads to the growth of oncogenic condition in different parts of the body.

Protein Kinases genes, like PIK3CA, because falls among those classes of “Cancer-related genes”, who normally stimulate cell growth but undergoing mutation, they become stimulated and highly active, which results into excessive cell growth and uncontrollable division of cell.

These types of mutations of PIK3CA gene, are found from the samples of patients of Breast, Ovary, Lung, Brain, Stomach, Liver, Colon (Colorectal Cancer), and, Rectum.

The Alteration pathways for formation of oncogenic condition in human body, through PI3K, due to PIK3CA gene are as follows:

  • Deletion of tumor suppressor PTEN (Phosphatase and TENs) gene
  • Amplification of regions in the gene which contain Akt
  • Onset of active point mutations at P13K

The most available disorder caused by the mutation of PIK3CA gene are Breast Cancer, ovarian cancer, brain cancer, colon cancer.

When should you consult your physician?

If you are facing prolonged symptoms like the above discussed (any of them), then you should definitely go for a medical consultation.

Diagnosis of PIK3CA mutation in human body

PIK3CA mutation results in the alteration of functioning of Protein Kinases and thus causing uncontrollable cell division, which leads to lumps in Breast cancer patients. Detection of mutation in PIK3CA gene is possible by Real TimePolymerase Chain Reaction (PCR).

Sample collection

It should be Biopsy sample, which can be tissue or fluid from breast lump.

What is Real Time-Polymerase Chain Reaction?

Real Time-PCR is a highly accurate and sensitive molecular biology technique to detect PIK3CA mutated gene in sample, where we can detect any type of somatic mutation in the sample gene.

Steps of Real Time-Polymerase Chain Reaction

  • Extraction: Sample bloods are treated with chemicals to have extracted or separated DNA from them.
  • Performing Polymerase Chain Reaction: Denaturation, Annealing, Extension or Amplification of the targeted gene of PIK3CA.
  • Repetition of the cycle: Total number of cycle for the process will be 40.
  • Time of report: The report of diagnosis with Real Time -PCR technique is 72 hours.

Obtaining result: The sensitivity and reproducibility of result is optimized using standards and the obtained record of amplification of concerned mutated gene PIK3CA, with whatever changes in the genetic base or mutation detected, will be analyzed by Molecular Biologist, and will be followed by final medical advisory by Doctor.

  • Indication of Positive and Negative Result: Any kind of mutation in the concerned gene PIK3CA  detected, will lead to positive result, and will be diagnosed as having mutated PIK3CA gene  as their reason for lump in breast which can be most possibly be diagnosed as malignant, whereas, it will be considered negative for detection of no mutated PIK3CA gene.
