Acid phosphatase tests are used to measure the level of acid phosphatase enzyme in your blood. This enzyme is found in various tissues of the body like Spleen, liver prostate gland etc. This test is primarily performed for the diagnosis of the certain medical conditions especially those related to the prostate gland

Historically Acid phosphatase test is used as a marker for the diagnosis of metastatic prostate cancer as elevated level of this enzyme is associated with this type of cancer

Now a days a more advanced techniques to detect prostate cancer is developed that is prostate specific antigen test(PSA)

It can also be used as a marker for detecting certain leukemias and specifically for many other diseases.A normal range of 2.5-3.7 ng/ml is good for our blood.

What is Acid Phosphatase Test?

What is Acid Phosphatase Test

  • Acid phosphatase is an enzyme that is found in the prostate, bone kidney Rbc etc.
  • The greatest concentration of it is found in the Prostate gland, spleen, liver bone marrow etc.
  • Majority of the prostate can be derived from RBC and prostate cells
  • The total concentration of it always increase during bone diseases

Sample Preparation

  • Take approx 5 ml of blood and centrifuge it for 2-4 minutes to get the clear serum
  • Use EDTA plasma because it stabilises the acid phosphatase
  • Avoid hemolysis
  • Morning sample is prefered
  • Immediately do the test
  • Acidify the serum below 6.5 for good stability

Acid Phosphatase Test Precaution

Acid Phosphatase Test Precaution

  •  Maintain temp of 37 c and ph below 7
  • It may have around 50 percent of its activity if kept at room temperature.

Moderately Raised Level Other Than Non Prostatic Tissue May Cause

  • Gaucher disease
  • Urinary retention
  • Myeloid leukaemia
  • Multiple myeloma
  • Sickle cell anaemia
  • Liver diseases like cirrhosis
  • Renal diseases

Important Facts:

  • Acid phosphatase level declines in 2-3 days after surgery
  • Acid phosphatase is not recommended to diagnose the prostatic carcinoma

Until Tumour Become Metastatic

  • Oestrogen therapy is also used to decline the level of acid phosphatase

Conclusion

Acid phosphatase test can be used for the the screening of prostate cancer however it is not always recommended but this test proves to be a very sensitive and specific test that should be conducted carefully and sample collection should be done carefully while doing test otherwise it may interfere with the results meanwhile the more advanced technique like PSA test has come that immediately diagnose the increased level of PSA due to its high specificity and sensitivity to the level of Acid phosphatase.
