What is IDH-1 test?

IDH stands for Isocitrate dehydrogenase. It is an enzyme that act as a catalyst in the oxidative decarboxylation of isocitrate leading to the formation of alpha-ketoglutarate and carbon dioxide (CO2).

Isocitrate is a component of citric acid cycle, also known as Krebs cycle. The reaction catalyzed by the IDH enzyme is usually irreversible and therefore it must be carefully regulated in order to:

  • Prevent depletion of Isocitrate
  • Avoid accumulation of alpha-ketoglutarate.

There are 3 isoforms of IDH in humans namely:

  • IDH 1
  • IDH 2
  • IDH 3

  • Affect the ability of T-cells to kill their target cells.

  • Your health care provider will first collect the samples such as whole blood and bone marrow aspirate in an EDTA tube.
  • Sample is sent to laboratory for further processing and testing.
  • After processing, DNA is extracted from the samples.
  • PCR is done to amplify DNA fragments containing IDH 1 and IDH-2 genes.
  • Later gene sequencing is done.
  • Reports are analyzed for IDH-1 and IDH-2 mutations.
